Naproxen is a non-steroidal anti-inflammatory drug (NSAID) with analgesic, antipyretic, and anti-inflammatory activity. The anti-inflammatory mechanism involves blockade of cyclo-oxygenase 1 and 2 in arachidonic acid metabolism, reducing prostaglandin synthesis. Its antiplatelet effect is related to inhibition of platelet cyclo-oxygenase, which reduces thromboxane concentration in platelets. Due to less efficacy than low-dose aspirin and inconsistency in clinical trials, naproxen is not used as a cardioprotective agent. The antipyretic effect is related to reduced prostaglandin PGE2 signaling on the hypothalamus.
⚠️ Warnings
Naproxen is registered as an over-the-counter medicine, creating a risk that patients may take multiple preparations containing the same active substance under different trade names. This increases the risk of adverse effects and gastrointestinal irritation. Due to elevated gastrointestinal bleeding risk, the lowest effective dose for the shortest duration is recommended.
