Coenzyme A(CoA,SHCoA,CoASH) is acoenzyme, notable for its role in thesynthesisandoxidationoffatty acids, and the oxidation ofpyruvatein thecitric acid cycle. Everygenomesequenced to date encodes enzymes that use coenzyme A as asubstrate, and around 4% of cellular enzymes use it (or athioester) as a substrate. In humans, CoA biosynthesis requirescysteine,pantothenate(vitamin B5), andadenosine triphosphate(ATP).

Acetyl-CoA is utilised in thepost-translational regulationandallosteric regulationofpyruvate dehydrogenaseandcarboxylaseto maintain and support the partition ofpyruvatesynthesis and degradation.
