Miconazole is an antifungal agent belonging to the azole class, used in the treatment of fungal infections. Its activity is based on inhibition of ergosterol synthesis through blockade of the fungal enzyme lanosterol 14α-demethylase. Miconazole is administered topically, typically twice daily. Possible adverse effects include local cutaneous reactions such as burning, erythema, and irritation. Miconazole is an inhibitor of CYP450 isoenzymes.
Miconazole (ATC D01AC02) exhibits antifungal activity against dermatophytes, yeasts, moulds, and dimorphic fungi. It also shows activity against certain bacteria. Its effects are mediated through inhibition of the fungal enzyme lanosterol 14α-demethylase.
This inhibits the synthesis of ergosterol from lanosterol, leading to accumulation of toxic precursors and disruption of fungal cell membrane formation.
