Olanzapine, sold under the brand nameZyprexaamong others, is anatypical antipsychoticprimarily used to treatschizophreniaandbipolar disorder.It is also sometimes usedoff-labelin cancer patients for treatment ofchemotherapy-induced nausea and vomitingand as anappetite stimulant.For schizophrenia, it can be used for both new-onset disease and long-term maintenance.It is takenby mouthor byinjection into a muscle.
Common side effects includefeeling tired,dizziness,constipation,dry mouth, andrestlessness.Other side effects includelow blood pressure with standing,allergic reactions,neuroleptic malignant syndrome,diabetes mellitus,seizures, andtardive dyskinesia.In older people withdementia, its use increases the risk of death.Use in the later part ofpregnancymay result in amovement disorderin the baby for some time after birth.Although its mechanism of action is not entirely clear, it is known to blockdopamineandserotonin receptors.
Olanzapine was patented in 1991 and approved for medical use in the United States in 1996.It is available as ageneric medication.In 2023, it was the 167th most commonly prescribed medication in the United States, with more than 3million prescriptions.It is on theWorld Health Organization's List of Essential Medicines.
⚠️ Warnings
• Caution should be exercised in patients with history of hypersensitivity to sulfonamides, liver or kidney impairment, seizures; children, pregnancy and breastfeeding.
• It may significantly increase blood pressure. Monitor blood pressure level regularly.
• It may cause drowsiness, do not drive a car or operate machinery while taking this medication.
Patient may develop with increased risk of Serotonin Syndrome with the following symptoms: increased heart rate, shivering, sweating, dilated pupils,etc . If it is so consult with your healthcare provider.
